本作叙述二大宗教佛教与基督教的创始者——佛祖和耶稣，两人在度过世纪末后到人间度假，在东京一起租了间公寓过生活的故事。剧情大多围绕在二人与现代社会的互动以及各种取材自佛教与基督教的题材元素上。

Saint☆Oniisan is a humorous drama about daily lives of Jesus Christ and Gautama Buddha who are living together as roommates in a Tokyo apartment while taking a vacation on Earth. The comedy often involves jokes about Christianity, Buddhism, and all things related, as well as the main characters' attempts to hide their identities and understand modern society in Japan.

世紀末を無事乗り越えたイエスとブッダは、しばしのバカンスを過ごすため下界にやって来た。東京・立川の安アパート・松田ハイツに部屋を借りた2人は、ルームシェアしながらのんびりとした日々を過ごす。厳しい修業時代の経験から、倹約家でお金に細かいブッダ、ついつい衝動買いしてしまうイエス。ともに聖なる神仏という立場にある一方、仕事を離れた下界では人間より人間くさい2人の生活は、小さな驚きや笑いにあふれていた。
