Op een vrachtschip worden er kernwapens gestolen door terroristen. Om een mogelijke Derde Wereldoorlog te voorkomen, worden Barney Ross, Lee Christmas en de andere leden van de Expendables ingeschakeld. Als 's werelds laatste verdedigingslinie trachten ze hier een stokje voor te steken met nieuwe teamleden die andere stijlen en tactieken toepassen.
