Larey Daley weet dat een Egyptisch tablet uit de tijd van de farao's het geheim is achter de tot leven komende museumstukken in het "Museum of Natural History". Maar wanneer dit magisch tablet tekenen van corrosie begint te vertonen, reist Larry met onder andere Farao Ahkmenrah, Teddy Roosevelt en Attila de Hun af naar het "British Museum" in Londen om het te redden van zijn ondergang.
Als het "Museum of Natural History" gesloten wordt vanwege renovaties, moeten de museumstukken overgebracht worden naar het beroemde "Washington Museum". Larry Daley (Ben Stiller) komt terecht in de zogenaamde Smithsonian-instelling van het museum, waar hij de opdracht krijgt om een aantal museumstukken te redden, omdat deze verkeerd zijn verscheept.
